Optimization of Tangential Flow Filtration (TFF) system in COVID-19 Vaccine Production in Hib Vaccine Production Division in PT Bio Farma, Bandung.
The appropriate course of action to stop the spread of COVID-19 involves
immunizations due to the rise in COVID-19 cases. Pichia pastoris serves as the protein
expression system for RBD, a spike protein produced by the SARS-CoV-2 virus. RBD is utilized
as an antigen for vaccine candidates to tackle COVID-19 which is suitable for low to
middle-income countries. The Tangential Flow Filtration (TFF) system is a method to
separate and purify molecules that are commonly used on the industrial scale. In this
project, the protein from the fermentation culture is successfully separated using the TFF
system, the RBD yield was 24.1 gr in batch 1, 26.8 gr in batch 2, and 30.2 gr in batch 3, and
43.9 gr in batch 4, achieving the target yield aimed by the PT Bio Farma. However, there are
limitations that cause protein loss during the process. Therefore, the issues are carried out
by reducing human error and optimizing the contact time between the buffer and
fermentation culture. Further research should be conducted regarding the production of
COVID-19 vaccine development on an industrial scale.
